IVF Laboratory Science Turns Attention to Denudation Protocols
EntSun News/11088796
LONDON - EntSun -- New discussion is emerging within the assisted reproduction community regarding oocyte denudation one of the most routinely performed yet least formally standardised steps in ICSI laboratory workflows.
Unlike culture media selection, incubator management, or vitrification, denudation technique has historically been passed down through training rather than governed by written evidence-based protocols. However, a growing body of peer-reviewed literature and internal audit data from progressive IVF units is beginning to challenge the assumption that this step is too simple to warrant formal scrutiny.
Key areas of concern include the duration of hyaluronidase exposure, the mechanical force applied during stripping, pipette sizing sequences, and the degree of technique variation between embryologists within the same laboratory. Each of these variables has been associated in published studies with downstream effects on spindle integrity, fertilisation rates, and blastocyst development.

Laboratories that have introduced standardised denudation protocols — including timed enzyme exposure, structured pipette sequencing, and competency-based training assessment report improvements in fertilisation consistency and reductions in oocyte degeneration rates.
The consensus view emerging from embryology conferences and quality improvement literature is that denudation deserves the same evidence-based framework now applied to other laboratory procedures.
